摘要内容:Bovineviraldiarrheavirus(BVDV)causesongoingeconomiclossestothelivestockindustry.Monitoringantibodiesviaenzyme-linkedimmunosorbentassay(ELISA)isakeytoolforensuringtheeradicationofBVDVfromcattleherds.WedevelopedanindirectELISA(rE2-iELISA)usingCHO-S-expressedrecombinantE2protein,themajorimmunogenicglycoproteinmediatingviralattachmentandimmuneevasion.Optimizedassayconditionsincluded:0.4mg/wellantigencoating,5%BSAblocking,1:100serumdilution,and1:5000secondaryantibodydilution.TheassaydemonstratedexclusivespecificityforBVDV-1andBVDV-2withdetectionsensitivityto1:1,500serumdilution.Validationrevealedexceptionaldiagnosticperformance:ROCanalysisshowed0.998AUC(cutoff=0.125),94.8%concordancewithIDEXXELISA,andtheintraand inter-batchcoefficientofvariationarebothlessthan5%.TheexperimentalresultsindicatethattheindirectELISAdetectionmethodbasedonBVDVrE2exhibitsgoodsensitivity,specificity,andstability.AndastableserologicaltoolforBVDVsurveillanceandvaccineefficacyevaluationincattlepopulations.
